环状RNA circTCF25在冠心病患者中的表达及临床意义

摘要
目的探讨环状RNA在冠心病(CAD)患者外周血单个核细胞(PBMCs)中的表达水平及临床意义。方法采用real-time PCR法检测CAD病例组和对照组PBMCs中circ TCF25、hsacirc005075和hsacirc1569的表达水平,利用Spearman相关分析探究circ TCF25与CAD危险因素及传统标志物的相关性,利用多元logistic回归分析及受试者操作特征分析(ROC)方法评估circ TCF25作为CAD生物标志物的可能性。应用低氧、TNF-α和IL-1β分别刺激人脐静脉内皮细胞24小时,检测circ TCF25的表达变化。结果与对照组相比,CAD患者中circ TCF25的表达水平显著降低(P<0.001),两组间hsacirc005075和hsacirc001569的表达水平无显著差异。circ TCF25的表达水平与空腹血糖、肌酸激酶和Gensini评分呈负相关(P<0.05)。circ TCF25的表达水平每增加一个单位,CAD发病风险降低45%(调整后OR值:0.55,95%CI:0.39-0.77,P<0.001)。circ TCF25的ROC曲线下面积为0.62(95%CI:0.57-0.67,P<0.001)。在低氧、TNF-α和IL-1β刺激内皮细胞24小时后,circ TCF25的表达水平均显著下降。结论 CAD患者PBMCs中circ TCF25表达水平显著下调,circ TCF25可能作为CAD潜在的生物标志物。
