IT-ISJ标记及其在陆地棉遗传图谱构建中的应用

摘要
【目的】开发可用于植物遗传多样性分析和遗传图谱构建的新型标记。【方法】根据植物结构基因内含子拼接位点的保守序列,设计扩增内含子的内含子-外显子拼接位点(intron targeted intron-exon splice junction,IT-ISJ)标记引物,并用于陆地棉遗传图谱构建。【结果】利用704个IT-ISJ引物组合,对陆地棉高品质品种渝棉1号和多显性基因标记系T586进行多态性筛选,获得49个多态性引物组合,占筛选引物的7.0%。49个多态性引物组合在(渝棉1号×T586)F2:7重组近交系群体270个系中检测到58个位点。以58个IT-ISJ、150个SSR和8个形态标记进行连锁分析,构建的遗传连锁图包括22个连锁群、113个位点(49个IT-ISJ、62个SSR和2个形态标记)。连锁图覆盖714.5cM,占棉花基因组的16.1%,标记间平均长度为6.3cM。【结论】IT-ISJ标记多态性高,可有效地用于陆地棉遗传连锁图谱的构建。
