河豚毒素单克隆抗体的制备及特性

河豚毒素(TTX)与钥孔(?)血蓝蛋白(KLH)结合生成完全抗原,用此抗原免疫BALB/C小鼠(5~6周龄),取2只小鼠脾细胞与SP2/O骨髓瘤细胞融合,ELISA法检测阳性克隆,共检出5株高效价抗TTX阳性克隆细胞,对此抗TTX单克隆抗体分析结果表明:(1)单抗对TTX有很高的抑制作用,10-9水平的毒素就可抑制单抗50%左右的A值。(2)抗体属IgG1亚类,IC50为2.5~5μmol/L。(3)抗体能阻断TTX引起的周围神经动作电位的抑制。(4)体内抗毒效果不如抗血清。
