天然产物来源多糖结构解析研究进展

摘要
多糖是高等植物、动物、微生物细胞中一类非常重要的天然大分子物质,由于许多活性多糖具有良好的免疫调节、抗肿瘤、抗氧化和调节肠道菌群等功能,因此科学认识多糖结构有利于深入研究其生物活性功能,对多糖的基础研究和应用具有重要意义。本文简述天然产物来源多糖分离纯化和结构解析的研究方法,重点介绍葡聚糖、葡甘聚糖、木葡聚糖、阿拉伯木聚糖和果胶等多糖结构解析的研究进展,就多糖结构解析目前存在的一些问题进行探讨与展望。通过归纳文献资料并结合笔者团队近20年的研究积累,提出"部分降解-甲基化-核磁共振"可作为多糖一级结构解析的核心技术,同时可辅助利用ESI-MS、Maldi-Tof-MS、离子淌度-质谱联用等技术与方法,从而构建一套先进的多糖一级结构解析技术体系。
