食物中毒菌多联融合毒素F5基因的构建及结构分析

摘要
目的:构建食物中毒菌多联融合毒素基因并分析其是否保持了原来毒素基因的抗原特性。方法:采用一定的linker序列(G-P-G-P-G)将克隆的3种食物中毒菌5个毒素基因片段进行串联,构建重组融合毒素Hc-VT1B-SEA-VT2B-SEB(命名为F5)基因,应用DNAstar,对F5融合蛋白抗原特性等进行预测分析。结果:构建的融合毒素F5保留了5种毒素的抗原特性,基因全长序列2 937 bp,编码成熟蛋白979个氨基酸,测序结果与设计序列(GeneBank)100%同源。结论:成功构建了食物中毒菌多联融合毒素F5基因,为下一步研究融合毒素F5的表达及利用融合毒素检测食物中毒菌毒素建立广谱免疫学检测新方法奠定了基础。
