腹腔注射干扰素γ对哮喘小鼠肺组织GATA-3及气道炎症的影响

摘要
目的:研究腹腔注射IFNγ对支气管哮喘的防治作用及其机制。方法:BALB/c小鼠随机分为三组,正常对照组(A组,12只)、哮喘模型组(B组,12只)、腹腔注射IFNγ组(C组,12只),B组、C组采用卵蛋白(OVA)和氢氧化铝建立哮喘模型,分别于第23天至第30天每次激发前腹腔注射0.25mlPBS液和IFNγ5μg,1次/d。第31天取肺泡灌洗液(BALF)测定细胞成分、IL4、IL5浓度,取血测定血浆IgE水平,观察肺组织病理学变化及GATA3的表达。结果:(1)C组BALF中的嗜酸粒细胞(EOS)(0.3±0.2)明显低于B组(21.1±6.7),P<0.01;(2)C组BALF中的IL4(3.0±2.5)和IL5(11.4±3.2)的水平明显低于B组的IL4(90.5±22.4)和IL5(55.6±10.7),P<0.01;(3)C组血浆中总IgE(24.3±7.6)和OVA特异性IgE(12.3±3.6)水平显著低于B组的IgE(67.2±9.7)和OVA特异性IgE(34.6±7.8),P<0.01;(4)HE染色示B组支气管平滑肌肥厚,黏膜充血、水肿,黏膜层增厚,并有EOS为主的炎性细胞浸润,管腔内可见黏液栓,支气管壁周围有EOS为主的炎性细胞浸润,而C组小鼠的上述炎性改变则明显减轻;(5)免疫组化显示B组肺组织中GATA3的表达明显增加,而C组GATA3的表达明显减少。结论:腹腔注射IFNγ可以抑制哮喘鼠IL4、IL5的合成、抑制EOS在气道内的炎性浸润及血浆中总IgE和OVA特异性的IgE水平,其机制可能与IFNγ阻断GATA3的表达,从而继发抑制Th2型反应有关。
