陈皮-半夏对内皮细胞损伤保护机制的研究

摘要：

目的研究陈皮-半夏能否通过对Beclin-1、微管相关蛋白1轻链3(LC-3)等自噬通路的调节作用及其空白干预内皮细胞损伤的机制。方法分离培养脐静脉内皮细胞(HUVEC),将其实验分为空白对照组,ox-LDL损伤模型组(模型组),陈皮-半夏小、中、大剂量组。MTT法,观察陈皮-半夏对内皮细胞存活率的影响;Western blot法观察陈皮-半夏对内皮细胞Beclin-1、LC-3表达的影响。方法与空白对照组比较,模型组细胞存活率显著下降(P<0.01),Beclin-1、LC-3表达显著升高(P<0.01)。陈皮-半夏中、大剂量组细胞存活率均较模型组显著上升(P<0.01),Beclin-1水平较模型组显著下降(P<0.01)。陈皮-半夏3个剂量组的LC-3水平均较模型组显著下降(P<0.05)。结论陈皮-半夏可能通过抑制血管内皮细胞中Beclin-1、LC-3的表达,发挥提高内皮细胞存活率治疗动脉粥样硬化的作用。
