氧化应激在胆汁淤积中的作用

摘要
<正>胆汁淤积是指胆汁形成和排泄障碍而引起的临床和生化紊乱综合征。多种原因都可以引起胆汁淤积,包括各类慢性肝病、药物、妊娠、感染、外科手术和Alagille综合征等。其中,原发性胆汁性肝硬化(PBC)和原发性硬化性胆管炎(PSC)是最常见的病因。毒性胆汁酸在血液和肝中的浓度升高,导致肝细胞损伤、炎症、胆管增殖和肝纤维化的发生。如果胆汁淤积情况未能得到纠正,纤维化最终进展为肝硬化和肝衰竭。不幸的是尚无治疗胆汁淤积的有效方法。一部分胆汁淤积患者可以使用熊去氧胆酸治疗(UDCA),一部分患者对糖皮质类固醇的抗炎药物反应良好。但是很多胆汁淤积患者缺乏好的治疗方法,唯有肝移植是一个可行的选择。因此,大量的研究针对胆汁淤积引起肝细胞损伤和纤维化的机制开展。其中许多研究聚焦在氧化应激在胆汁淤积时肝细胞损伤中的作用。一、胆汁淤积动物模型中的氧化应激活性氧簇(ROS)在每个有氧代谢的细胞中持续产生。细
