吴茱萸碱对人胃癌SGC7901细胞增殖、侵袭和保罗样激酶-1基因表达的影响

摘要
目的:观察中草药提取物吴茱萸碱(evodiamine)对人胃癌细胞侵袭的影响及可能的机制。方法:采用不同浓度的吴茱萸碱处理人胃癌SGC7901细胞后,以软琼脂集落培养试验检测癌细胞锚着不依赖性增殖,以Boyden小室模型方法检测癌细胞侵袭能力;分别以荧光实时定量RT-PCR和免疫荧光方法检测癌细胞保罗样激酶-1(polo-likekinase 1,PLK1)基因表达水平。结果:胃癌SGC7901细胞经吴茱萸碱处理后,恶性增殖和穿膜细胞数均明显下降,且呈剂量依赖性(P<0.05)。吴茱萸碱处理组PLK1 mRNA和蛋白水平均明显下调,且呈时间和浓度依赖性(P<0.05)。结论:吴茱萸碱可明显抑制胃癌细胞侵袭力,其机制可能与下调PLK1基因表达有关。
