降香新黄酮latifolin通过Nrf2/HO-1通路抗H9c2细胞缺氧复氧损伤作用研究

摘要
目的:研究降香新黄酮latifolin对心肌H9c2细胞缺氧复氧损伤的保护作用及其可能机制。方法:建立H9c2细胞缺氧复氧损伤模型,实验分正常对照组、模型组及不同浓度latifolin(2.5、5、10、20μg/mL)组;采用MTT法检测细胞活力,微量酶标法测定细胞上清LDH活性;荧光染色法和流式细胞仪分析细胞中ROS含量;免疫荧光染色法和Western blot检测胞浆中Nrf2、HO-1及胞核中Nrf2蛋白表达。结果:与模型组比较,latifolin能显著升高H9c2细胞活力,降低细胞上清中LDH及细胞中ROS水平,升高胞浆中Nrf2、HO-1及胞核中Nrf2蛋白表达(P<0.05)。结论:latifolin对H9c2细胞缺氧复氧所致损伤具有保护作用,其机制可能与激活Nrf2/HO-1通路有关。
