针刺天枢穴对结肠炎肠上皮细胞NF-κB信号通路的影响

摘要
目的:研究针刺"天枢穴"对DSS诱导结肠炎(IBD)的治疗作用及其对核因子-κB(NF-κB)信号通路的影响。方法:将30只C57BL/6小鼠随机分为空白组、模型组和天枢穴组,模型组和天枢穴组分别给予3%DSS水溶液造模,天枢穴组每日给予20min针刺干预,连续7d。记录每日DAI评分,取材后测量结肠长度,HE染色观察结肠病理变化,ELISA法检测结肠组织白介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)水平,Western blot法检测结肠组织中NF-κB、IκBα蛋白磷酸化水平,免疫荧光检测结肠组织中NF-κB表达水平。结果:天枢穴组每日DAI评分显著低于模型组(P<0.01);结肠长度针刺组显著长于模型组(P<0.05);结肠组织HE染色观察发现天枢穴组炎性损伤明显轻于模型组;结肠组织中IL-6、TNF-α含量天枢穴组显著低于模型组(P<0.01),NF-κB、IκBα磷酸化水平亦明显低于模型组(P<0.01)。结论:针刺天枢穴可以明显地改善DSS诱导IBD症状及结肠组织炎性反应损伤程度,可能是通过抑制NF-κB信号通路而起到了治疗作用,针刺天枢穴可以作为IBD一种有效的治疗手段。
