大蒜素对白假丝酵母菌生物被膜的抑制作用

摘要
目的研究大蒜素对白假丝酵母菌生物被膜(BF)内真菌的抑制作用。方法用FITC-conA标记白假丝酵母菌BF的多糖成分,利用荧光显微镜从形态学上观察大蒜素对白假丝酵母菌BF形成过程的影响。将标本分为5组,分别是对照组(大蒜素浓度为0)、大蒜素0.25mg/mL组、大蒜素0.5mg/mL组,大蒜素1.0mg/mL组、大蒜素2.0mg/mL组。应用XTT减低法定量检测大蒜素对白假丝酵母菌BF中真菌的杀灭作用。结果在荧光显微镜下可见大蒜素2.0mg/mL组大蒜素干预白假丝酵母菌BF后的荧光强度较大蒜素0.5mg/mL组及对照组的荧光强度减弱。大蒜素0.5、1.0、2.0mg/mL作用于白假丝酵母菌BF 5h后,XTT减低法光密度(OD)450的值分别为1.87±0.31、1.64±0.25和1.30±0.29,分别与对照组的2.11±0.26比较,差异均有统计学意义(P<0.05);0.25mg/mL的大蒜素作用白假丝酵母菌BF后,OD450值为1.93±0.31,与对照组比较,差异无统计学意义(P>0.05)。结论大蒜素能杀灭白假丝酵母菌BF内的真菌,对BF有抑制作用。在一定浓度范围内,随着大蒜素浓度的增加,其对BF的抑制作用逐渐增强。
