miR-21靶向调控STAT3基因抑制人乳腺癌MCF-7细胞的侵袭

摘要
目的探讨miR-21和信号传导与转录激活因子3(signal transducer and activator of transcription 3,STAT3)基因在人乳腺癌MCF-7细胞中的表达,阐明miR-21对STAT3基因的靶向作用及其对MCF-7细胞侵袭的影响。方法购买并培养MCF-7细胞,采用免疫荧光法检测STAT3在癌细胞中的表达。运用生物信息学方法对miR-21和STAT3基因的靶向配对关系进行预测,采用荧光素酶报告系统鉴定;脂质体2000转染miR-21模拟物后,qRT-PCR检测miR-21和STAT3 mRNA在癌细胞中的表达,Western blot检测STAT3蛋白在癌细胞中的表达,Transwell小室检测MCF-7细胞体外的侵袭性。结果光镜下可见人乳腺癌MCF-7细胞成片生长,有突起;细胞免疫荧光法检测结果显示胞质内有STAT3蛋白的表达,显示红色荧光。生物信息学软件miRanda和Target Scan显示miR-21和STAT3基因二者靶向配对良好,荧光素酶报告系统鉴定发现miR-21 mimics能够抑制STAT3 mRNA表达。qRTPCR和Western blot检测结果表明过表达miR-21能够降低STAT3 mRNA和蛋白的表达。Transwell小室实验结果表明miR-21的过表达能够抑制MCF-7细胞的侵袭。结论 miR-21通过负性调控人乳腺癌MCF-7细胞中STAT3基因的表达,进而抑制癌细胞的侵袭。
