基于含药血清经时谱效关系的半夏白术天麻汤抑制细胞焦亡的药效物质研究

摘要
目的:运用含药血清经时谱效关系研究经典名方半夏白术天麻汤保护内皮细胞免受棕榈酸诱导细胞焦亡的药效物质。方法:建立棕榈酸诱导的人脐静脉内皮细胞(HUVEC)焦亡模型,以细胞活力、一氧化氮(NO)、丙二醛(MDA)、总超氧化物歧化酶(T-SOD),抑制其释放炎性介质白介素-6(IL-6)、肿瘤坏死因子α(TNF-α)作为半夏白术天麻汤含药血清抑制细胞焦亡的药效学评价指标,建立大鼠灌胃后吸收进入血化学成分的分析方法,采用典型相关性的谱效关系分析方法指认半夏白术天麻汤抑制细胞焦亡的有效成分。结果:柚皮苷、大豆苷元、甘草素、异甘草素和白术内酯Ⅲ与抑制细胞焦亡的正相关性较大,可明显增强细胞活力,改善MDA、NO、T-SOD、IL-6、TNF-α水平。结论:半夏白术天麻汤中橘红、甘草、白术的成分参与保护棕榈酸诱导的HUVEC细胞焦亡,含药血清经时谱效关系可用于发现半夏白术天麻汤的血管内皮细胞保护成分。
