肿瘤患者髓源抑制性细胞的免疫抑制及临床应用分析

摘要
髓源抑制性细胞(MDSCs)是处于不同分化阶段的异质性、未成熟髓系细胞,对维持肿瘤患者免疫逃逸和耐受,促进肿瘤生长、增殖及转移等方面具有重要作用。由于缺少特异性标志物及MDSCs异构性,人MDSCs表型尚未完全明确。人MDSCs能够通过上调关键酶降解氨基酸、诱导氧化应激、辅助性与调节性T细胞、激活PD-1/PD-L1轴、诱导高丰度肠道真菌及高表达抑制性细胞因子等机制抑制免疫应答,作为肿瘤预测标志物和治疗靶点应用于临床。检测外周血、腹腔液、肿瘤组织中MDSCs有助于肿瘤临床诊疗和预后判断,通过抑制MDSCs功能、促进其分化为更成熟的细胞、破坏细胞内在结构链等可降低MDSCs的比例和表型。本文就国内外关于MDSCs临床应用相关分析的研究进展,探讨其可能存在的免疫抑制机制并分析其临床应用现状,从MDSCs入手改善肿瘤微环境免疫抑制状态,有望为肿瘤免疫治疗提供新途径。
