经NF-κB通路调节骨桥蛋白表达对骨关节炎滑膜细胞炎症影响

摘要
目的探讨经核因子kappa B(NF-κB)通路调节骨桥蛋白(OPN)表达对人膝关节滑膜炎的影响。方法收集20例原发性膝关节骨关节炎(OA)病人滑膜标本(OA组),以因膝关节内骨折行急症手术、半月板或韧带损伤3d内手术病人滑膜标本15例作为对照组。采用免疫组化法观察两组膝关节滑膜中OPN及NF-κB通路相关蛋白的表达;体外分离培养获取成纤维滑膜细胞,采用Western blot方法检测膝关节成纤维滑膜细胞中OPN及NF-κB通路中p65、磷酸化p65(p-p65)、IκBα和磷酸化IκBα(p-IκBα)的表达。结果与对照组比较,OA组滑膜炎症明显。免疫组化显示,OA组滑膜组织中OPN表达明显高于对照组,NF-κB信号通路激活,差异有统计学意义(t=4.796~16.980,P<0.05)。Western blot检测显示,OA组成纤维滑膜细胞中OPN、p65、p-p65、p-IκBα表达较对照组显著增加,差异有统计学意义(t=3.351~16.980,P<0.05);而两组通路抑制蛋白IκBα表达差异无显著性(P>0.05)。结论 OA滑膜增生与炎症可能与NF-κB信号通路激活介导的OPN的表达有关。
