滤泡辅助性T细胞及Th9在可诱导共刺激分子基因敲除小鼠类风湿关节炎模型中的作用

摘要
目的探讨可诱导共刺激分子(inducible costimulator, ICOS)敲除基因小鼠类风湿关节炎(rheumatoid arthritis, RA)模型中ICOS信号对于滤泡辅助性T细胞(follicular helper T cells, Tfh)和Th9细胞的影响。方法流式细胞仪分析测定ICOS敲除基因(ICOS-knockout, ICOS-ko)小鼠及对照组小鼠Tfh及Th9细胞群的表达;酶联免疫吸附实验(enzyme linked immunosorbent assay, ELISA)检测脾淋巴细胞培养悬液中细胞因子中白细胞介素(interleukin, IL)-21、IL-9的水平。结果野生型RA小鼠脾细胞Tfh亚群表达水平从8 W到16 W呈上升趋势[(8 W:(2.55±0.95)%;16 W:(15.30±3.48)%];ICOS-ko RA小鼠脾细胞Tfh亚群的表达同样呈上升趋势[8 W:(0.91±0.49)%;16 W:(2.56±1.03)%], 但与野生型RA小鼠相比各期均表达下调(均P<0.05);野生型RA小鼠脾细胞Th9亚群表达水平从8周到16周呈上升趋势[8 W:(2.57±1.20)%;16 W:(8.93±2.56)%];ICOS-ko RA小鼠脾细胞Th9亚群的表达同样呈上升趋势[8 W:(0.94±0.49)%;16 W:(2.58±1.32)%], 但与野生型RA小鼠相比各期均表达下调(均P<0.05);野生型RA小鼠的IL-21、IL-9的表达从8周到16周呈上升趋势, ICOS-ko RA小鼠同野生型RA小鼠相比, 表达趋势相同但呈下调趋势(P<0.05)。结论 Tfh细胞及Th9细胞参与了RA的免疫应答过程;下调ICOS能够下调Tfh及Th9的致炎作用。
