低聚半乳糖对应激大鼠肠黏膜屏障功能的影响

摘要
目的研究肠道补充低聚半乳糖(galacto-oligosaccharides,GOS)对冷束缚应激(cold restrain stress,CRS)大鼠肠黏膜紧密连接和屏障功能的影响。方法 40只Wistar大鼠随机分成正常对照组、GOS组、CRS组和CRS+GOS组,每组10只,正常对照组仅给予基础饮食,GOS组每天给予GOS(4g/kg)喂养,共10d;CRS组大鼠于普通喂养第10天,被束缚固定后置于4℃冰箱3h,制备CRS模型;CRS+GOS组在每天给予GOS(4g/kg)喂养的第10天制备CRS模型。建模成功后,各组大鼠经心脏采血,检测血浆二胺氧化酶(diamine oxidas,DAO)浓度;处死大鼠,取回肠组织行病理检查及电镜检查,再采用免疫组化法检测细胞紧密连接蛋白occludin的表达,并以实时定量PCR法检测其在mRNA水平的表达。结果与正常对照组相比,CRS组及CRS+GOS组大鼠血浆DAO浓度均明显增高(P<0.01);CRS组血浆DAO浓度又明显高于CRS+GOS组(P<0.01);CRS组及CRS+GOS组大鼠肠黏膜occludin在蛋白和mRNA水平的表达均较正常对照组显著降低(P<0.01);CRS组上述指标又明显低于CRS+GOS组(P<0.01),正常对照组与GOS组间上述指标的差异均无统计学意义(P>0.05)。结论添加GOS喂养可以降低大鼠血浆DAO浓度,增加回肠occludin在mRNA和蛋白水平的表达,提示GOS可能具有改善紧密连接和维护肠屏障功能的作用。
