参芪解郁方对产后抑郁大鼠脑前额叶皮层,海马区5-HT,NE及其受体的干预机制

摘要
目的:从脑前额叶皮层,海马5-羟色胺(5-HT),去甲肾上腺素(NE)及其受体变化角度,探讨具有健脾养心作用参芪解郁方对产后抑郁症(PPD)的干预机制。方法:选用雌性SD大鼠,2~3月龄,随机分为正常、假手术、模型、中药、西药组,每组分设1,2,4,6周观察时点。采用去势后连续雌孕激素注射并突然停撤方法制备PPD模型,并给予药物干预。规定时点取材,选脑前额叶皮层、海马区域,采用高效液相法测定5-HT,NE含量,免疫组化法检测5-羟色胺受体(5-HT1AR),去甲肾上腺素受体(α2AR)蛋白表达。结果:(1)模型组各时点前额叶皮层、海马5-HT,NE含量均较正常、假手术组显著减少(P<0.01)。与模型组2,4,6周比较,中药组、西药组前额叶皮层、海马5-HT,NE含量增加(P<0.05,P<0.01)。(2)与正常、假手术组各时点比较,模型组前额叶皮层、海马5-HT1AR蛋白表达显著减少(P<0.01),α2AR蛋白表达显著增强(P<0.01);与模型组2,4,6周比较,中药组前额叶皮层及海马5-HT1AR蛋白表达增加(P<0.01);中药组2周及中、西药组4,6周前额叶皮层α2AR蛋白表达减少,中药组4,6周,西药组2,4,6周海马α2AR蛋白表达减少(P<0.05,P<0.01)。结论:参芪解郁方可明显提升PPD大鼠脑内5-HT,NE含量,并能对其受体表达的异常状态进行有效调整,对上述神经递质及其受体的平衡调节可能是该方发挥抗抑郁效应的重要途径之一。
