应用ATR-FTIR研究二种皮肤渗透促进剂的作用机理

应用衰减全反射傅立叶红外光谱(ATR -FTIR)为检测手段 ,通过测定人体在体皮肤表面不同深度角质层的CH2 - 收缩振动峰的波数位移和角质层中脂质的分布,来考察二种常用皮肤渗透促进剂油酸(OA)和月桂氮卓酮(Azone)对角质层固有脂质的影响。以探索渗透促进剂在人体皮肤角质层中的作用机理。二种渗透促进剂均使得νasCH2-和νsCH2-发生蓝移 ,但Azone的作用更强。OA和Azone对皮肤角质层深部的脂质含量无明显影响。由于角质层最外层吸附了一定量的OA ,致使皮肤最外层的脂质含量的升高。Azone同样也只是吸附在角质层外层 ,并未大量进入角质层深部。就CH2-收缩振动峰的波数位移而言 ,OA和Azone均与人体整个角质层中细胞间的脂质发生作用 ,使得脂质侧链的无序性变大 ,导致了流动性的增大 ,进而减少了药物透皮吸收的阻力。
