金龟子部分种类COI基因序列比较分析

摘要
对金龟总科(Scarabaeoidea)6种金龟子线粒体细胞色素氧化酶I(COI)基因部分序列进行了PCR扩增和序列测定,并从GenBank中检索4个种的COI基因序列,比较其同源性并获得了长度为624bpCOI片段。10个种的COI序列分析结果表明:该基因序列共有440个变异位点,192个简约信息位点,表现出明显的A+T偏倚(63.6%)。转换(TV)和颠换(TS)在科内、科间存在明显差异。科内种间TV+TS小,各科不同种与外群之间TV+TS较大;TV/TS则呈现相反的趋势。以黑蜣科(Passalidae)的具角黑艳甲(Odontotaenius disjunctus)作为外群,采用P-distance法重建了10种金龟子邻接树(N-J)和最小进化树(ME)拓扑结构,结果表明:丽金龟属内科内、鳃金龟科内的不同种分别优先聚集,再与其他科相聚集,显示科内、属内的种间有较近的亲缘关系,说明COI基因序列可用来研究属内或科内种群的系统发育问题。
