microRNA-34a在大鼠膝骨关节炎模型中的功能研究

摘要
目的探讨微小RNA-34a(miR-34a)在碘乙酸钠所致大鼠膝骨关节炎模型中的表达变化,并初步明确其功能。方法选取30只雄性Wistar大鼠,左侧膝盖不注射碘乙酸钠作为对照组,右侧膝盖注射碘乙酸钠作为模型组;造模完成后检测两组大鼠关节液内miR-34a表达水平的变化;分离两组大鼠的膝关节软骨细胞并进行原代培养,敲低miR-34a后,使用流式细胞仪测定细胞凋亡情况。结果模型组Makin评分为(5.09±1.35)分,对照组为(1.27±0.64)分,差异有统计学意义(P<0.05)。与对照组比较,模型组中miR-34a在关节液和关节软骨细胞内的表达均升高,差异均有统计学意义(P<0.05)。将miR-34a用它的反义互补片段敲低后,模型组大鼠关节软骨细胞的凋亡受到明显抑制(P<0.05)。结论 miR-34a在碘乙酸钠所致大鼠膝骨关节炎的关节液中表达升高,并且抑制miR-34a的表达会降低关节炎大鼠的关节软骨细胞凋亡率。
