基于蛋白质结构的药物分子设计

摘要
随着蛋白质晶体学和分子模型技术的发展,基于蛋白质结构的药物分子设计已成为药物设计的主要潮流之一。这类方法从与疾病有关的蛋白质的三维结构出发,利用计算机程序,先对药物结合位点进行描述,然后通过各种方法产生潜在的配体分子,最后才配体分子进行打分,给出设计结果。目前已出现了多种基于蛋白质结构的药物分子设计方法,也有了一些商业化的软件。文章综述了近年来出现的有关设计方法,列举了成功的设计实例,并对各种方法的优缺点以及所存在的问题进行了简要的讨论。
