白藜芦醇对小鼠移植性肝癌组织中细胞周期蛋白的影响

摘要
目的 研究白藜芦醇 (Res)对小鼠移植性肝癌组织中细胞周期蛋白CyclinD1,CyclinB1和p34cdc2表达的影响 ,探讨Res抗肝癌作用及其机制 .方法 利用小鼠移植性肝癌H2 2模型研究Res的体内抗肿瘤效果 ,免疫组织化学检测实验组与对照组小鼠肝癌及瘤旁肝组织CyclinD1,CyclinB1和p34cdc2蛋白的表达情况 .结果 Res治疗组能不同程度抑制小鼠肝脏移植瘤的生长 ,10mg·kg- 1 和 15mg·kg- 1 的Res治疗时抑瘤率分别为 36 .3%和 4 9.3% ,与对照组比较有显著差异 (P <0 .0 5 ) .与对照组相比 ,Res治疗组癌组织中CyclinD1的表达无明显变化 (6 6 .7%vs 6 0 .0 % ,P >0 .0 5 ) ,而CyclinB1(77.8%vs30 .0 % ,P <0 .0 5 )和p34cdc2 (88.9%vs 4 0 .0 % ,P <0 .0 5 )的表达明显降低 .结论 Res具有抗肝癌作用 .通过抑制Cy clinB1和p34cdc2的表达影响细胞周期的进程 ,可能是Res抗癌作用的机制之一 .
