脓毒症大鼠肾脏线粒体膜电位变化和超微结构损伤的研究

摘要
目的探讨脓毒症时大鼠肾脏线粒体的损伤情况。方法采用腹腔注射内毒素(LPS)建立脓毒症大鼠模型。30只SD大鼠随机分为对照组、6 h脓毒症组和24 h脓毒症组。分别检测各组大鼠血清肌酐(Cr)、尿素氮(BUN)水平;电镜观察肾脏线粒体形态学变化;运用流式细胞术检测分离肾脏线粒体的肿胀程度和线粒体膜电位,用前向角与侧向角的平均荧光强度比值(FSC/SSC)反映线粒体肿胀程度,用二通道和一通道平均荧光强度比值(FL2/FL1)确定线粒体膜电位。结果脓毒症大鼠的血清Cr、BUN上升明显高于对照组(P<0.05);肾脏线粒体肿胀程度(FSC/SSC)在24 h脓毒症组中明显增加(对照组:0.55±0.10;6 h脓毒症组:0.58±0.10;24 h脓毒症组:0.66±0.12,P<0.05);肾脏线粒体膜电位(FL2/FL1)在6 h脓毒症和24 h脓毒症组中均明显下降(对照组:0.77±0.26;6 h脓毒症组: 0.32±0.19;24 h脓毒症组:0.30±0.17,P<0.01),其下降趋势与大鼠血清Cr变化呈负相关(r= -0.510,P=0.004);肾脏线粒体超微结构在24 h脓毒症组中损伤明显。结论脓毒症早期肾脏功能和线粒体膜电位存在损伤;但线粒体超微结构损伤在脓毒症早期表现轻微,晚期损伤明显。
