损伤动脉外膜重塑和成纤维细胞表型的变化

摘要
观察兔腹主动脉损伤后血管外膜及外膜细胞增殖和表型特征的变化 ,采用病理形态学、计算机图像分析、免疫组织化学和透射电镜方法观察兔腹主动脉损伤后血管外膜和外膜细胞的变化。结果发现 ,球囊损伤后 3天、7天、14天和 2 8天血管外膜厚度均显著增加。外膜细胞密度在损伤后 3天开始增加 ,7天显著增加。外膜细胞增殖指数在损伤后 3天达到高峰 ,7天仍显著增高。外膜细胞的α actin染色反应在损伤后 3天由阴性转为弱阳性 ,7天和 14天呈强阳性。损伤后 7天和 14天外膜细胞大量微丝和粗面内质网明显扩张 ,呈现出肌成纤维细胞的特征。结果提示 ,动脉损伤后早期血管外膜显著增厚 ,外膜细胞增殖活跃 ,成纤维细胞逐渐转化为肌成纤维细胞 ,伴有收缩蛋白合成增加 ,这些变化在病理性血管重塑中发挥重要作用
