即刻早期基因c-fos,c-jun在缺血再灌注大鼠肝脏中的表达

摘要
目的研究即刻早期基因家族中c-fos、c-jun基因在缺血再灌注肝脏中的表达情况,探讨其与缺血再灌注损伤发生的关系。方法 SD大鼠随机分为假手术组、缺血再灌注后30min组和缺血再灌注后120min组。全自动生化分析仪测定血清ALT、AST水平,光镜观察肝组织形态学改变,TUNEL法测定肝细胞凋亡指数。实时荧光定量PCR(realtimePCR)法测定组织中c-fos及c-jun的表达情况。结果肝脏缺血再灌注损伤后血清ALT、AST逐步上升(P<0.05)。病理改变显著,细胞凋亡现象逐步显现。c-fos及c-jun的表达水平,在再灌注后30min明显升高(P<0.05),再灌注2h后表达水平有所回落,但仍显著高于假手术组(P<0.05)。结论 c-fos及c-jun基因参与肝脏缺血再灌注损伤的发生,并可能与肝细胞凋亡与再生的过程相关。
