葡萄糖、游离脂肪酸对培养的人血管内皮细胞一氧化氮含量的影响

摘要
目的 观察不同浓度的葡萄糖 (Glu)、软脂酸 (PA)和油酸 (OA)对培养 2 4小时的人血管内皮细胞一氧化氮 (NO)生成的影响。方法 采用硝酸还原酶法。结果  Glu(30 mmol/ L )组和 Glu 30 mmol/ L +PA0 .2 5 mmol/ L组细胞上清液中 NO含量明显高于对照组 (P<0 .0 5 ) ;OA(0 .2 5、0 .5 mmol/ L )组 ,PA+OA(PA 0 .2 5 mmol/ L +OA0 .5 mmol/ L )组及 Glu+PA+OA (Glu 30 mmol/ L +PA 0 .2 5 mmol/ L +OA 0 .5 mmol/ L )组细胞上清液中 NO含量明显低于对照组 (P<0 .0 5 ) ;PA (0 .12 5、0 .2 5、0 .5 mmol/ L )组与对照组比较 ,上清液中 NO含量无明显差异 (P>0 .0 5 )。结论 高浓度 Glu作用于内皮细胞 2 4小时使内皮细胞生成 NO增多 ,而 OA抑制内皮细胞生成 NO。高浓度 OA与高浓度 Glu联合作用时 ,OA抵消了 Glu的作用 ,仍然使 NO降低。高浓度 Glu和游离脂肪酸对内皮细胞 NO生成出现的相反效应的机制应进一步研究
