脑脉泰对大鼠局灶性脑缺血再灌注损伤的抗炎保护作用

摘要
目的研究脑脉泰对大鼠局灶性脑缺血再灌注损伤缺血区炎症反应的影响。方法大脑中动脉线拴法(MCAO)制作大鼠局灶性脑缺血再灌注模型:雄性SD大鼠随机分为假手术组(sham)、脑缺血再灌注模型组(MCAO)、脑脉泰大、中、小剂量组(MCAO+脑脉泰2.24、1.12、0.56g/kg)和尼莫地平组(MCAO+尼莫地平10mg/kg)。用TUNEL法检测缺血半暗带凋亡细胞,用免疫组化染色法检测NF-κB、TNF-α、ICAM-1和IL-1β的蛋白表达水平。结果脑缺血再灌注损伤明显增加MPO活性和NF-κB、TNF-α、ICAM-1和IL-1β的表达,应用不同剂量的脑脉泰后,上述效应明显减弱,同时,凋亡细胞也明显减少(P<0.01)。结论脑脉泰减少脑缺血/再灌注损伤,其保护作用与抑制MPO活性和降低NF-κB、TNF-α、ICAM-1、IL-1β的表达,进而抑制炎症反应有关。
