CD14/TLR-4-NF-κB信号通路参与骨关节炎发病机制的研究

摘要
目的观察膝骨关节CD14、toll样受体(toll-like receptor,TLR)-4、NF-κBp65、基质金属蛋白酶(matrix metallo-proteinases,MMPs)-13蛋白及其mRNA表达,探讨膝骨关节炎发病机制。方法 40只清洁级雄性SD大鼠,按体重随机分为两组。正常组同步饲养,模型组采用改良Hulth法建立膝骨关节炎模型。第4周末处死全部大鼠,摘取膝关节滑膜组织,应用HE染色观察膝骨关节组织形态学变化;分别应用Real-time PCR、Western-blot技术检测CD14、TLR-4、NF-κBp65、MMPs-13mRNA及其蛋白表达。结果模型组CD14、TLR-4、NF-κBp65、MMPs-13mRNA及其蛋白表达呈高表达特点,与正常组比较有统计学意义(P<0.05)。结论 CD14/TLR-4-NF-κB信号通路过度活化,下游NF-κBp65及MMPs-13的表达升高可能是导致关节软骨破坏,降解加速,诱发骨关节炎发展的重要因素之一。
