不同剂量姜黄素对宫颈癌裸鼠移植瘤生长及瘤组织MIF、VEGF表达的影响

摘要
目的探讨不同剂量姜黄素对宫颈癌裸鼠移植瘤生长及瘤组织巨噬细胞移动抑制因子(MIF)、血管内皮生长因子(VEGF)表达的影响。方法建立人宫颈癌Caski细胞裸鼠移植瘤动物模型,按随机数字表法将动物模型分为五组:阴性对照组(羧甲基纤维素钠)、顺铂组(3 mg·kg-1·d-1)、姜黄素低剂量组(50 mg·kg-1·d-1)、中剂量组(100 mg·kg-1·d-1)和高剂量组(200 mg·kg-1·d-1),每组7只。阴性对照组给予羧甲基纤维素钠灌胃15 d,顺铂组每隔3 d腹腔注射给药,共5次,姜黄素组连续灌胃给药15 d。停药24 h后处死各组裸鼠,测量裸鼠体重,剥取瘤体,测量移植瘤体积及瘤重,计算抑瘤率。采用实时定量PCR(RT-PCR)试验和免疫组化法分别检测各组瘤组织中MIF、VEGF mRNA和蛋白的表达情况。结果顺铂组和姜黄素低、中、高剂量组的移植瘤体积均明显小于阴性对照组(均P<0.05),不同剂量姜黄素组抑瘤率两两比较差异均有统计学意义(P<0.05);RT-PCR结果显示,与阴性对照组相比,顺铂组、姜黄素低、中、高剂量组瘤组织中MIF、VEGF mRNA的相对表达量均显著减少(均P<0.05);MIF mRNA相对表达量在姜黄素低、中、高剂量组之间差异无统计学意义(P>0.05);姜黄素低剂量组的VEGF mRNA相对表达量与姜黄素中、高剂量组及顺铂组相比差异均有统计学意义(P<0.05);免疫组化结果显示:顺铂组、姜黄素低、中、高剂量组瘤组织中VEGF、MIF蛋白表达明显低于阴性对照组(均P<0.05),姜黄素低剂量组的VEGF蛋白相对表达量与姜黄素中、高剂量组及顺铂组相比差异均有统计学意义(P<0.05)。结论姜黄素对宫颈癌裸鼠移植瘤的生长具有明显的抑制作用,姜黄素能够降低瘤组织中MIF、VEGF的表达水平。
