调肝方药对愤怒、郁怒模型大鼠下丘脑5-HTR2C基因表达的影响

摘要
目的:通过研究愤怒和郁怒模型大鼠下丘脑5-HTR2C基因的表达,探讨愤怒和郁怒的微观发生机制及中药经前平颗粒、经前舒颗粒的作用靶点。方法:居住入侵法制备愤怒和郁怒证模型大鼠,经前平颗粒、经前舒颗粒干预,均10 g·kg-1,ig,分别为正常组、模型组、经前舒组、经前平组;运用半定量RT-PCR和Western blot方法检测大鼠下丘脑区5-HTR2C mRNA及其蛋白表达水平。结果:与正常组相比较,愤怒组和郁怒组大鼠下丘脑中5-HTR2C mRNA和蛋白相对水平均极显著性降低(P<0.01);愤怒组大鼠的相对表达水平改变明显强于郁怒组大鼠;用药后,与模型组相比较,5-HTR2C mRNA均得到显著性改善,但与正常组相比仍极显著性下降(P<0.01),而蛋白改变均得到极显著性改善,与正常组相比无统计学差异。结论:模型组大鼠下丘脑与5-HTR2C基因表达极显著降低,调肝方药干预可显著上调模型大鼠下丘脑5-HTR2C mRNA和蛋白表达,说明5-HTR2C基因与愤怒和郁怒证相关,并且可能是经前平颗粒和经前舒颗粒治疗该证的作用机制之一。
