摘要
The aim of the present study was to investigate the development of buccal mucoadhesive tablets for local delivery of chlorhexidine diacetate, an antimicrobial drug largely used for treatment of periodontal disease. Buccal tablets were prepared with mesoporous silicate MCM-41 in combination with a mucoadhesive polymer to study the influence of mesoporous material incorporation on mucoadhesive characteristics, drug delivery, and antifungal activity. Chlorhexidine diacetate adsorption on the MCM-41 pore surface was performed and the inclusion/adsorption product was characterized. Mucoadhesive tablets were then prepared by using different mixtures of hydroxyethylcellulose/MCM-41 and sodium carboxymethylcellulose/MCM-41. All tablets were characterized for their swelling, ex vivo mucoadhesive time and ex vivo mucoadhesive force. Lastly, in vitro drug release and antifungal activity were evaluated on chlorhexidine diacetate-loaded tablets. Tablets made a 2:1 ratio of hydroxyethylcellulose/MCM-41 showed the best mucoadhesive properties, good drug release, and good antifungal activity.
