Recent crystal structures of the T-cell receptor (TCR) engaged with peptide-major histocompatibility complex (MHC) ligands suggest the TCR binds to such ligands in a conserved orientation, with key V alpha residues contacting the alpha helices. TCR mutagenesis has permitted energy measurements of these interactions. Here, Thomas Manning and David Kranz review binding energetics data that might explain the inherent reactivity of TCRs with MHC products.
