Angiogenesis inhibition: Drugs in clinical trials

Under physiologic conditions, in most adults angiogenesis is required for wound healing and maturation only. In malignancies, angiogenesis is required for the growth of solid tumors and the development of metastases. The heterogeneity of tumors does not influence this process. Complete inhibition of angiogenesis is targeted towards the endothelial cells of growing capillaries and should therefore be well tolerated. Anti-angiogenic agents with different modes of action are prepared for or are in clinical trials. Among these drugs are TNP-470, thalidomide, CM101,carboxyamido-triazole, matrix metallo-proteinases inhibitors, vascular endothelial growth factor targeting compounds, isotretionin, interferon-alpha, tamoxifen, tecogalan combrestatin, and squalamine. First reports from these trials give interesting information about the pharmacokinetics, response rates, and side effects. These data are summarized in this review article.
