TRPV1 expression extends to a number of cells besides cutaneous afferents, suggesting functions for this molecule in diverse sensory and non-sensory processes. Possible side effects related to visceral, thermoregulatory, cognitive, motor, or emotional functions must therefore be considered during the development of therapeutic strategies involving the systemic administration of agents active at TRPV1. Proper dosing and biodistribution will undoubtedly be critical parameters in these strategies, as in others aimed at molecules presumed to be 'nociceptor-specific'.
