Galanin in the hypothalamus raises dopamine and lowers acetylcholine release in the nucleus accumbens: a possible mechanism for hypothalamic initiation of feeding behavior

Rats were prepared with two implanted guide shafts, one for microdialysis to measure extracellular dopamine (DA) and acetylcholine (ACh) in the posterior, medial nucleus accumbens (NAc), and the other for microinjection of galanin, neuropeptide Y or saline in the hypothalamic paraventricular nucleus (PVN). There was an increase in DA release and a decrease in ACh in the NAc following microinjections of galanin into the PVN. The effect was observed only in rats for which identical galanin injections induced feeding in separate tests. Ringer injections had no effects. Unlike galanin, neuropeptide Y in the PVN induced eating without altering DA/ACh; whereas earlier results showed that norepinephrine in the PVN works like galanin. These results suggest that galanin initiates feeding, in part, by activating the mesolimbic DA system and suppressing intrinsic cholinergic activity in the NAc. This may prime instrumental behavior with DA while disinhibiting behavior by lowering ACh. (C) 1998 Elsevier Science B.V. All rights reserved.
