摘要：

PI3K is a key regulatory node in growth- factor signaling across evolution. Its importance in a wide array of physiologic processes, including insulin signaling, cell growth, immunity, and brain development, has been confirmed in animal models and humans. In particular, PI3K has held great promise as an anticancer target from the time of its initial discovery. Multiple targeted agents are now available, and we expect that they will be proved effective when coupled with strategies to minimize insulin feedback.
