One-step fabrication of a bienzyme glucose sensor based on glucose oxidase and peroxidase immobilized onto a poly(pyrrole) modified glassy carbon electrode

A one-step and all-electrochemical procedure for the fabrication of a bienzyme based amperometric biosensor is reported. Horseradish peroxidase (HRP) and glucose oxidase are co-immobilized onto a glassy carbon electrode in an electropolymerized polypyrrole (PPy) film. The hydrogen peroxide produced by the oxidase can be detected through its HRP mediated reduction in a potential window (0-200 mV vs. Ag/AgCl) ensuring the electrical conductivity of the film and minimising electroactive interferences. PPy films of different thickness are employed to modulate the sensor linearity and sensitivity. The operational characteristics of the bienzyme sensor are described.
