FET based biosensor for estimation of the effect of Ca2+ and Mg2+ on the immobilised butyrylcholinesterase

摘要
The activity of immobilised butyrylcholinesterase (BChE) was determined potentiometrically by field-effect-transistor (FET), using butyrylthiocholine iodide (BTCh) as a substrate. The enzyme activity increased in the presence of Ca2+ and Mg2+. Computation of experimental data showed: (1) the increase of enzyme and substrate affinity in the presence of Mg2+; and (2) the increase of the rate of formation of hydrolysis products from the enzyme-substrate complex in the presence of Ca2+ and Mg2+ Positive cooperativity was observed for Mg2+, the Hill coefficient increased from 1.23 to 2.37 with the increase of Mg2+ concentration from 0.1 to 10 mM. For Ca2+ the coefficient was about 1.0 independently from the concentration. (C) 1998 Elsevier Science S.A. All rights reserved.
