Novel approach to study the initial events in the folding and assembly of procollagen

摘要
The folding and assembly of procollagen occurs within the endoplasmic reticulum through a series of discrete steps which eventually lead to the transport of a trimeric, triple helical molecule to the Golgi apparatus. Our understanding of the temporal and spatial organization of this maturation process has been enhanced by studying the assembly and secretion of both wild-type and mutated molecules using cells grown in culture. More recently cell-free systems have been developed which reconstitute the assembly process as they would occur in the intact cells. This enables a number of fundamental questions to be addressed concerning the molecular recognition events occurring during the assembly of this molecule. (C) 1996 Academic Press Ltd
