In this issue of Cancer Cell, Saha et al. systematically test and optimize combination therapy strategies in a challenging model of glioblastoma. Durable complete responses were seen only when an oncolytic virus expressing IL1 2 was coupled with anti-CTLA-4 and anti-PD-1 therapeutics.
