Sugar transport through channels reconstituted in planar lipid membranes

We have reconstituted maltoporin, a maltose-specific transmembrane protein, into planar lipid membranes and recorded the power spectrum of the ion current after application of a small transmembrane voltage. The presence of maltose in the aqueous phase causes blocking of the channels in a concentration-dependent manner. The power spectra of the ion current show, in addition to the 1/f dependence, a pronounced Lorentzian with a corner frequency proportional to the maltose concentration. A series of measurements with different maltose concentrations yields the on and off rate of maltose to the channel. Under simplified conditions this allows an estimate of sugar translation.
