DNA fragmentation attested by TUNEL in Saccharomyces cerevisiae subjected to photodynamic treatment

Terminal deoxynucleotide transferase-mediated dUTP nick end labelling (TUNEL) was adapted to the detection of DNA double strand breaks in yeast cells subjected to photodynamic treatment using hematoporphyrin as a photosensitizer. This DNA fragmentation appears early during the treatment and increases dramatically after the plasma membrane becomes permeable to propidium iodide. These results suggest that the loss of plasma membrane integrity is not the primary cause of yeast cell inactivation, as previously reported. (C) 1997 Elsevier Science B.V.
