Effect of nanometer hydroxyapatite particles on rat macrophage at mRNA level

摘要
To investigate the potential cytotoxicity of nanometre hydroxyapatite (nano-HA), the transcription and expression of genes about DNA damage were determined by semi-quantitative RT-PCR. The results showed that 100 mu g/ml nano-HA can induce P53, P21 and HSP70 up-regulated, and these genes have obviously difference compared with negative control (p < 0.05). Furthermore, their expressions were depending on the concentration of particles. But the influence of nano-HA on Gadd45 was not visible. Above results indicate that cytotoxicity of nano-HA particles on macrophage would be determined by these genes at mRNA level, which can regulate cell cycle and DNA damage. Moreover, P53 and HSP70 would be ideal indictors to evaluate the biological safety of nanometer ceramics particles.
