Oxygen availability and ABA metabolism in Fagus sylvatica seeds

At harvest, beechnuts have a very deep dormancy which lies both in the structures surrounding the embryo and within the embryo itself. Covering structures prevent embryo germination by interfering with water uptake and gaseous exchange. To understand the role of the covering structures and oxygen availability on ABA catabolism, (+)-[H-3] ABA metabolism was studied in isolated embryos as well as in intact seeds. ABA degradation resulted essentially in oxidative products (PA, DPA). These products were more abundant in isolated embryos than in intact seeds. They accumulated mainly as alkali-nonhydrolyzable conjugates of DPA. A small amount of free and esterified forms were often observed. In isolated embryos a decrease in oxidative products was observed either by lowering the oxygen availability or by feeding embryos with tetcyclasis (an inhibitor of monooxygenase). In the presence of the covering structures, these oxidative products were reduced in the same manner, indicating that covering structures were probably responsible for limiting the oxygen supply to the embryo and for the low germination percentage observed in the case of intact seeds.
