The optimization of in vivo nuclear polyhedrosis virus production in Spodoptera exempta (Walker) and Spodoptera exigua (Hubner)

Production of the nuclear polyhedrosis viruses (NPV) of Spodoptera exigua and S. exempta was studied experimentally to determine the conditions for optimum in vivo production in the homologous hosts. Larvae of S. exgua and S. exempta were separately reared in individual cells on artificial diet which had been sprayed with a range of concentrations equivalent to 1 x 10(2) to 1 x 10(7) occlusion bodies (OB)/cell. The maximum yield obtained for individual larvae was 1.40 +/- 0.28 x 10(9) OB (+/-SE) for S. exigua dosed as 5-15 mg larvae with 1 x 10(6) OB/cell and harvested after 5 days' incubation and 3.29 +/- 0.46 x 10(9) OB for S. exempta dosed as 31-50 mg larvae also with 1 x 10(6) OB/cell and harvested after 7 days' incubation. Optimized yields per diet cell were 8.02 +/- 2.40 x 10(8) OB for S. exigua and 2.30 +/- 0.59 x 10(9) OB for S. exempta. LT50 values fell with dose from 10.3 days at 1 x 10(4) OB/cell to 5.5 days at 1 x 10(7) OB/cell for S. exempta. Postharvest incubation led to slight OB yield increases for S. exigua but not for S. exempta and to a large increase in bacterial contamination. Low to moderate OB doses increased larval weight relative to that of control larvae in both species. (C) 1997 Academic Press.
