Detection of plum bark necrosis stem pitting-associated virus (PBNSPaV) from different stone fruit species and optimisation of diagnostic tools

摘要
Stone fruit trees, showing clear-cut symptoms of stem pitting, were assayed by RT-PCR for the presence of PBNSPaV. A DNA fragment of the expected size (290bp) was amplified from total nucleic acid extracts from cortical scrapings of almond 'Filippo Ceo', peach 'Springcrest' and 'May Crest', plum 'Black Beaut', apricot 'Castelbrite' and 'Tyrinthos', and sweet cherry 'Ferrovia'. The test was repeated monthly throughout year using different tissues (flowers, leaves, dormant phloem) to identify the optimal sampling period and tissue type for reliable diagnosis. Different protocols for total nucleic acid extraction and RT-PCR amplification were compared.
