SOLUBLE HEMAGGLUTININS OF ADENOVIRUSES BELONGING TO ROSENS SUBGROUP-3 .2. SLOWLY SEDIMENTING HEMAGGLUTININ

Zonal centrifugation of soluble components of adenovirus types 1, 2, 5 and 6 revealed five different components that represented, listed in sequence of decreasing sedimentation rate, penton aggregates (rapidly sedimenting complete HA; RHA), hexons, isolated pentons, slowly sedimenting complete HA (SHA) and fibers. SHA eluted immediately after fibers and distinctly before pentons in anion exchange chromatography. It differed from penton-associated complete HA by the facts that it (a) did not carry any toxin activity, (b) was resistant to trypsin treatment, (c) was unaffected by prolonged heating and incubation with a moderate concentration of guanidin-HCl and (d) did not increase in HA activity in the presence of heterotypic antisera. A comparison of the behaviour of SHA in zonal centrifugation and in exclusion chromatography suggested it to be a markedly elongated structure. It is assumed to represent an aggregate of two fibers. This together with the observations on penton and capsid associated HA of adenoviruses belonging to subgroup III indicates that possession of at least two erythrocyte reactive sites is a prerequisite for any of the structures to exert hemagglutinating activity. © 1969 Springer-Verlag.
