ANTIOXIDANT ENZYMES ASSOCIATED WITH THE BLOOD-CELLS AND HEMOLYMPH OF THE MUSSEL MYTILUS-EDULIS

Immunocytochemical localisation of the antioxidant enzymes catalase, superoxide dismutase (SOD, both the Cu/Zn and the Mn form) and glutathione peroxidase (GPX) was carried out on blood cells from the mussel Mytilus edulis at both the light and electron microscope level. The results showed the SOD (Cu/Zn) antibody localised in association with the plasma membrane of the hyalinocytes. The granular haemocytes gave a positive reaction for all the antibodies investigated. Catalase was localised in small electron dense granules indicating that these granules are peroxisomes. Both forms of the SOD enzyme were associated with granules, with the labelling density generally lower for the Mn form. Biochemical measurements of antioxidant enzyme activities supported the immunocytochemical results. Catalase and SOD activities were present in blood cells but not detectable in haemolymph. The SOD activity was indicated to be due to both Cu/Zn and Mn forms of the enzyme (respectively, 75·9 and 24·1% of total activity). GPX activity was present in the cells but predominantly in the haemolymph (87·5% of total). © 1993 Academic Press. All rights reserved.
